ABSTRACT Purpose: As has been quite well known to the field of psychiatry for some time, people who end up in trouble with the law often do so because of severe mental illness. Sometimes this mental illness can take the form of a personality disorder, which at present is considered incurable, or it can take the form of severe depression, for which there are currently many treatments. Dna Replication DNA replication is a complex cellular function that is necessary in order to sustain life and achieve growth. Many enzymes, proteins, and other molecules work together to ensure that genetic information is replicated efficiently, quickly, and accurately. Without any one of these components, replication would be very limited in its efficacy. DNA is comprised of two strands of complementary nitrogenous bases (adenine & thymine, guanine & cytosine), five-carbon sugars (either ribose or deoxyribose), and phosphate groups. The strands of DNA are arranged in a double-helix array and are held together with hydrogen bonds. The semiconservative replication model is used to depict replication. In this model, each new double helix has one old strand and one new strand. This is yet another way in which accuracy is ensured. Because the shape of the DNA molecule is extremely important to its functionality, care must be taken to ensure that all parts of the molecule remain in their appropriate space during replication, and that no part of the strand is broken. To replicate DNA, the two strands must first be separated from one another. The first enzyme used in this process is called helicase. Helicases use the energy from ATP molecules to unwind the three-dimensional double helix. While the strand is unwinding, topoisomerase enzymes (such as gyrase) prevent the strands from being winded into a supercoil due to the torque produced by the separating action. Since each strand is comprised of complementary base pairs that have a high affinity to hydrogen-bond with one another, single-stranded binding proteins (SSBs) are attached to the strands to keep them from reattaching to one another. Once the strands are separated, work can begin to construct two new complementary strands that will ultimately attach to the existing DNA strands to form new complete DNA sequences. DNA polymerase III is the active enzyme that builds the new complementary strands. DNA polymerase III is a DNA-dependent enzyme. As such, a template (the existing separated strand) must be present to generate the new strand. DNA polymerase III requires a primer to begin its action. The primer used is a short RNA sequence with a 3' hydroxyl group that is formed by an enzyme known as primase. This primer is usually about ten nucleotides in length and is complementary to the existing DNA strand. DNA polymerase always works in the same direction: from the 5' end to the 3' end. Since DNA polymerase III always works in the 5' to 3' direction, and DNA strands are complementary, this gives rise to a few minor issues that must be dealt with. The strand in which DNA polymerase can move in the same direction as gyrase (with the replication fork) is known as the leading strand. As the strand is unwound, DNA polymerase III can easily begin to replicate the strand, as the replication fork is already moving in the 5' to 3' direction. The complementary strand is known as the lagging strand. The replication fork is necessarily moving in the 3' to 5' direction on this strand. On this strand, numerous primer sequences are inserted so that the DNA polymerase III can backtrack to build the new sequence as the strand is unwound. The DNA sequences between these primers, which are 1000 to 2000 nucleotides long, are known as Okazaki fragments. Once DNA polymerase III has replicated the fragments, the need arises to remove the RNA primer sequences and fuse the portions of the new strand together. The first critical enzyme used to do this is known as DNA polymerase I. This enzyme removes the primer sequence with the crucial 3' hydroxyl group and synthesizes complementary DNA to fill in the gaps left by the primers. After this is completed, yet another enzyme known as ligase is used to join the fragments. This enzyme works by forming a phosphodiester bond between the 3' hydroxyl of the new strand and the 5' phosphate group found on the Okazaki fragment. At the start of the book Estella is very mean and insulting to Pip. This is not her own nature, and that is why it is possible for her to change into a better person. Estella was brought up by Miss Havisham to crush the hearts of all men, so that is what she does to Pip. Since Miss Havisham is the only family Estella has ever known, she is bound to her. Since she is bound to Miss Havisham, she is obligated to serve as her tool of vengeance. But, when Miss Havisham dies, that bond is gone and more of Estellas true nature can come out. Since Estella is, at heart, a well-meaning person, she does soften up. This occurs after she has experienced hard times as Bentley Drummles wife, wich forces her to convert, and her cruel personality is finally destroyed by her true nature. At the end of the book Pip realizes that she has changed and thinks: ...what I had never seen before, was the saddened softened light of the once proud eyes; what I had never felt before, was the friendly touch of the once insensible hand. White culottes and this sexy pink sweater† (p. 86) The importance of this character at the beginning is that she is sweet and innocent, and her clean pink and white clothes are a sharp contrast to the dirty green uniforms that everyone wears on the battlefield. Her boyfriend Mark Fossie is supposed to have arranged for her to come over to join him, and no doubt this is the dream of every soldier. The whole episode is obviously fantasy and wish fulfilment. The story of Mary Ann Bell’s corruption by the horrors of war is symbolized in her appearance when she is found in the creepy cave. The author stresses that she is the same person that she was before, an d yet something dreadful has happened to her: â€Å"She was barefoot. She wore her pink sweater and a white blouse and a simple cotton skirt.† (p. 105) This cheerleader image is shattered, however with the revelation: â€Å"At the girl’s throat was a necklace of human tongues.† (p. ... The innocent eyes of girls at home are contrasted with the fictitious Mary Anne Bell who did share the boys’s experience: â€Å"She was there. She was up to her eyeballs in it.† (p. 108) The legend of Mary Anne Bell is the legend of the girlfriend who turned into a better soldier than all of the young men and actually enjoyed the terror and the gruesome body parts and awful smells. Mary Anne Bell is both the idealized dream girlfriend, and the terrifying nightmare girlfriend who loses her cuteness and becomes just as depraved and hardened as the men. Neither version is a real person, of course. It seems that the men need these psychological extremes to love and to hate. Mary Anne Bell is a harmless target of their strongest emotions and thinking about her helps them to work through difficult feelings. Some of the male characters pretend that they are great lovers who can attract many women, and this betrays an attitude of seeing women as sex objects. An example of this is the stories that Rat Kiley told about his exploits: â€Å"If Rat told you, for example, that he’d slept with four girls one night, you could figure it was about a girl and a half.† (page 85). All of the women characters in the book are presented through the eyes of the soldiers and they are all to some extent used by the soldiers as objects. The scene in the hotel where Biff finds out about Willy’s other woman devastates him, and ruins his life later on. Due to this, he decides not to attend the summer school and finish his studies. Considering his actions, Willy can be blamed for Biff’s inability to reach his dreams. The boy’s mindset is very much affected by his father’s actions. In contrast to Obama Sr. who serves his country and continues his studies, Willy shares in the disappointment of his son. These examples show us the important role of the father in giving advice to their sons, and in setting a good example. Despite the fact that Obama leaves his son to his wife, the good memory he leaves the boy serves as a good foundation for the child. Moreover, as the older Obama expresses in one of his letters to his son, for one to realize his place in society, one has to know one’s people, where he belongs (66). To do this, Barry searches for his roots and listens to what his grandmother’s story. From this, he finds out the struggles of his grandfather and his father, and finds strength in their sacrifices and dreams. The dreams of his father to be fully educated and to serve his country are enough to inspire Barry to move on and be the best he can be. In contrast, the theme of knowing oneself in order to realize one’s dreams is expressed differently in â€Å"Death of a Salesman. † In the play, Willy’s high expectations of Biff disappoint the son more. Biff insists to his father, â€Å"Pop, I’m nothing! I’m nothing, Pop. Can’t you understand that? There’s no spite in it any more (99)† Despite this, Willy still believes in Biff. He believes that if given a chance and a big sum to start on his own, his son can be successful. This is the reason why Willy sacrifices his life at the end, so that his sons can have what it takes to start on their own. The legacies of the two fathers are totally different. Obama leaves to his son an identity and reputation he can live with, while Loman bequeathes a good sum, which he is unable to provide his sons during his lifetime. The purpose of this study is to examine the impact of brand image and advertisement on consumer buying behavior in the general public at Gujranwala city. Questionnaire survey was used to collect the data by using non probability convenient sampling technique. A sample of 200 questionnaires was used in which 175 responses were collected within the period of one month. Findings show that brand image and advertisement have strong positive influence and significant relationship with Consumer buying behavior. People perceive the brand image with positive attitude. Study depicted that teenagers in Gujranwala are more conscious about their social status so they prefer branded products and advertisement affects their Consumer Buying Behavior positively. The Strengths and Weaknesses of Augustine’s and Ireneaus Theodicy ‘Evil did not come from God, since God’s creation was faultless and perfect’ (St Augustine) St Augustine was a man wrote a theodicy about the problem of evil and suffering based on the Creation and Fall of Genesis in the bible, he believed that evil was caused by something that was outside the omnipotence of God. God himself is all powerful but after creating the world, he Himself would not be able to control the actions that human beings create in changing it. When God created the world, with absolutely no evil and suffering, he saw what he had done and saw it was all good. God did not want evil he wanted humans and angels to enjoy the world the way that he created it to be. Evil is said to have come from the off spring of Adam, who was the first human on earth. Augustine wrote that Satan, a fallen angel, had tempted Adam and Eve with the power and free will of the earth when God had completed the creation of it. This temptation was proved to much for Adam and Eve who wanted to have power over the earth, this was one of the starter points for evil. In the bible there is said to have been an angel called Lucifer, this angel was among the other angels. He was the ‘Lord of Light’. However, he turned to evil and fell in to the darkness. This darkness is said to have evolved Lucifer in to Satan. Satan lives in hell, and hell is the darkness of all evil. Suffering has evolved from the consequences of evil, evil had started from the small part of greed of humans and had effected the world as human beings and angels each began to disobey God.
